Prop Alasdair Dickinson also has a shoulder worry, with Richie Gray, John Barclay, Rory Lamont and Hugo Southwell are all giving cause for concern.
Scrum half Rory Lawson should be fit after missing out on Saturday.
Given the casualty list, Scotland coach Andy Robinson has delayed naming his side for the Springboks challenge until Wednesday morning and may be considering a few changes after a record defeat at the weekend.
Second row Gray, who made his first international start against the All Blacks after three replacement appearances, is unsure whether he has done enough to retain his starting spot.
“I was happy with some things, disappointed with others,” he said.
I was happy with some of my set-piece work, a couple of good spells in attack. I got to grips with the game, but I’ll need to improve a lot.
“The physicality and the pace of the game is something I’ll need to improve and adapt to.
“We let ourselves down and the nation down.
“However, this is the positive about the autumn Tests. We’ve got another game to bounce back and get ourselves back on track.”
If the 21-year-old recovers from his dead leg and keeps his place, he could face the formidable Springboks second row combination of Bakkies Botha and Victor Matfield.
“The challenges just get bigger and bigger, but that’s international rugby,” added the Glasgow Warriors player.
“We’ll build this week and we’ll rectify the areas that went wrong.
“We’ll remain positive and, on the Saturday, we’ll have done everything to go out and put on a performance.”
South Africa arrive in Edinburgh having ground out narrow wins over Ireland and Wales on their autumn travels.
This is going to a real tough one (for Scotland, don’t think Dickinson, Rory Lamont and Grey will be such a big loss normally but given the siutation with the injuries Scotland already have its starting to look a bit thin. The others listed will definitely be missed and in my starting line up when all are available, but at least Rory Lawson will be a boost at scrumhalf if he is fully fit. Will be interesting to see who finally makes the team.

I’m convinced that there is now a bit of potency in the Bokke backline.
Hope they cut out the needless kicking game and respect the possession the Bokke gets.
Apart from outside center, the players are playing their natural postions.
